A study on growth ability of Thymus vulgaris L. under impact of chemical and physical factors of culture medium

Thyme plants (Thymus vulgaris L.) belonging to the family Lamiaceae have been used as ingredients in food processing and drugs in treating respiratory, gastrointestinal or nervous disorders, thanks to the secondary metabolites, such as thymol and carvacrol, existing in its leafy essential oils. However, the formation and accumulation of thyme oils are mostly affected by the variation of environmental and ecological systems. This study aimed to find appropriate conditions for the in vtro growth of thyme plants under stable and controlled temperature (T 24 ± 2oC) and relative humidity (RH 50 ± 5%).

On day 28, the number of shoots was the largest (4.3 shoots/explants) when nodal explants containing dormant buds were cultured on the MS medium suplemented with 1 mg L-1 BA, 0,5 mg L-1 IBA and 30 g L-1 sucrose under PPF of 40 µmol m-2 s-1 and photoperiod of 12 h d-1. When cultured photoautotrophically (on sugar-free medium), in vitro thyme plants derived from nodal cuttings grew significantly on MS salt medium having macro-elements 1/2 and supplemented with 200 mg L-1 KNO3 and 200 mg L-1 KH2PO4 under the PPF of 95 µmol m-2 s-1 and photoperiod of 12 h d-1 on day 35.
