High frequency shoot regeneration of dark blue statice (Limonium sinuatum L. Mill) by combining phytohormones with adenine in in vitro culture

Abstract

In the purpose of optimizing the micropropagation of dark blue Statice (Limonium sinuatum L. Mill), the appropriate age of explants, the effects of phytohormones, GA3 and adenine on Statice shooting were studied. MS medium (Murashige and Skoog, 1962) with 100 mg/l myo-inositol, 30 g/l saccharose, 6 g/l agar was used in all experiments. The cultures were in 16h photoperiod, light intensity of 2800 lux and at a temperature of 25 ± 2ºC. The best suitable explants were shoots from six-week seedlings in vitro. In the influence of cytokinin, 4 mg/l kinetin was the best result for statice shooting (5.6 shoots per explant) after 8 weeks of culture. The number of shoots per explant on MS medium supplemented with 4 mg/l kinetin and auxin (IBA, NAA or 2,4-D) was lower than MS medium with 4 mg/l kinetin but the size of shoots was raised. The presence of GA3/adenine increased shoot sprouting. The combination of 4 mg/l kinetin with 10 mg/l GA3 or 10 mg/l adenine raised the number of shoots per explant (7.8 and 9.73 shoots per explant, respectively) after 8 weeks of culture.
