Study on acclimatization methods of Dianthus caryophyllus L. in vitro

Dianthus caryophyllus L. is one of the world’s most popular cut flower. Market demand for carnation is rising, therefore, the propagation methods have also been researched. In this study, some acclimatization methods were investigated. At first, shoot tips were cultured on MS medium supplemented with different concentrations of BA and NAA. The highest response percentage was occurred when explants cultured on MS medium supplemented with 3.0 mgl-1 BA and 0.2 mgl-1 NAA (86.19%). Although, there were not statistic significant difference with combination of 2.0 mgl-1 BA and 0.2 mgl-1 NAA (75.51%), 2.0 mgl-1 BA and 1.0 mgl-1 NAA (82.08%). The highest multiplication rate (5.18 shoots/explant and 4.86 shoots/explant) were obtained by culturing the explants on MS medium supplemented with 2.0 mgl-1 BA and 1.0 mgl-1 NAA, 3.0 mgl-1 BA and 1.0 mgl-1 NAA. After that, containers: nylon bag, cyclindrical flask, erlen flask were used to study growth of carnation shoots. Shoots grew in nylon bags, have 2 air transfer holes than others under light density of 80 µmol m-2s-1, light regime of 12 hours per day. Osram lights stimulated growth of carnation plantlets. The best result was occurred when shoots were cultured under Osram violet light system (ppfd: 55 µmol m-2s-1) or Osram fluorescent light system (ppfd: 75 µmol m-2s-1).
