Optimization of culture conditions for coq10 biosynthesis by recombinant Agrobacterium tumefaciens

Abstract

Coenzyme Q10 (CoQ10) is increasingly being used as the functional foods, improving the health; furthermore CoQ10 also can be used in treatment of several severe diseases such as cardiovascular disease, diabetes, Parkinson… and added to cosmetic as an anti-aging agent for skin. Therefore CoQ10 demand is greatly increased. Production of CoQ10 can be performed by several ways such as chemical synthesis, semi-chemical synthesis and biotechnology, in which biosynthesis is more advantages and increasingly prioritized. In the present study, some optimal culture conditions for CoQ10 producing recombinant Agrobacterium tumefaciens DPXS12 strain were investigated. For investigated sugars (sucrose, lactose, glucose, galactose, mannose, xylose), the highest level of CoQ10 was obtained from A. tumefaciens DPXS12 using 5% sucrose as carbon source. Corn steep liquor (1%) in combination with (NH4)2SO4 (2%) was an appropriate nitrogen source for CoQ10 production by A. tumefaciens DPXS12. Optimal conditions for CoQ10 production by recombinant A. tumefaciens DPXS12 were 28oC and pH 7.0. The growth and CoQ10 production dynamics indicated that the highest CoQ10 level was 96 h fermentation.
